Using this method a specific immunochemical reaction between antibodies and antigen is employed to analyse a corresponding compound within a biological matix.

The assay is based on competition between the drug conjugate and any free drug to bind with an antibody. If a drug is present in the specimen undergoing analysis, it competes with drug conjugate for antibody-dye conjugate, which is in limited supply. If the quantity of drug is equal to, or greater than, the cut-off levels of any of the drugs, it inhibits binding of the drug conjugate to the antibody.

This is why, as illustrated below, no test line is visible when a result is positive whereas the presence of a coloured band indicates a negative result.

Procedure


Each ToxSure Oral II drug test kit contains of the following inclusions:

  • Untreated sample collector
  • Buffer bottle
  • Test device

The following instructions provide details on how to use ToxSure II to accurately analyse a saliva sample for drugs of abuse.

  • Remove test device from foil pouch and ensure each testing chamber contains purple dye.
  • Instruct the donor to rub swab on the inside of their cheek and around gums for 15 seconds on each side of the mouth.
  • The collector is to insert the swab into buffer solution and rotate 10 times. Remove swab, Place the dropper lid on to buffer bottle.
  • Dispense 7 drops of the buffer solution into each testing chamber. Close the lid of each chamber softly until lid clicks into place. DO NOT PUSH THE LID FULLY CLOSED.
  • Shake test devise vigorously for approximately 10 seconds and observe pinkish colour of the solution. Start timer for 4 minutes.
  • After 4 minutes, close the lid completely by pushing down firmly until lid snaps into place, piercing membrane beneath chambers. Shake test device from side to side for 3 seconds.
  • Observe pink migration in results window.
  • Results may be interpreted from 6 minutes if both C lines are present. If one or more of the Test Lines are absent, results may be read in the 11th minute. Do not read results after 12 minutes.

Interpretation

Negative

The presence of a line next to both the Control “C” and test indicates a negative result. Please note that this does not necessarily mean there is a complete absence of drug in the specimen, only that the level is below the cut-off given in Australian standard AS 4760:2006.

Positive

The absence of a line next to the test and the appearance of a line next to the Control “C” indicates a positive test result. As with a negative result, this is an indication the concentration of tested drug(s) in the specimen is above the particular cut-off level of that component.

Invalid

If there is an absence of a line in the Control “C” area following the alloted time the test should be considered invalid. In this case the sample should be re-analysed with a new test, in strict accordance with the documented procedure.

Interpretation of drug test results

Note: It is important to understand that the colour intensity of Test “T” lines can vary and that the presence of any line, no matter how faint, is still indicitive of what the test suggests.

The fuel cell within the FC10 has two 1″ platinum electrodes which hold an acid-electrolyte compound between them. As the test subject exhales, air flows over one side of the cell, which causes any alcohol present to be oxidised into acetic acid and also produce protons and electrons.

The electrons pass via a wire from the platinum electrode to an ammeter which measure the current and then on to the platinum electrode on the other side. After migrating through the lower portion of the fuel cell, protons, oxygen and electrons combine on the other side, forming water and carbon dioxide. Thus, the greater the quantity of alcohol that is oxidized during this process, the greater the electrical current, which can be measured and converted by a microprocessor into an acurate value for Blood Alcohol Concentration (BAC).

Procedure


Each FC10 alcohol breath test device kit contains of the following inclusions:

  • 1 Year warranty

The following instructions provide details on how to use the FC10 to conduct an automatic test in order to determine the blood alcohol level of a test subject.

  • Turn on the FC10 using the power button.
  • Verify the display reads “AUTO TEST.”
  • Attach an EasyTab™ mouthpiece to the back of the unit.
  • Instruct subject to blow into the mouthpiece ?rmly and steadily for as long as possible
  • Read the result.
  • Following a test, results are displayed on screen.
  • Press the Function button to return to the test mode.

Note: If the total expired volume is less than 1.3 litres an error message is displayed and the test must be conducted again.

Interpretation

1. Breath Flow

As the subject blows into the mouthpiece the FC10 displays a graphical representation of the breath flow on the display, in addition to the volume (in litres) in the upper left corner.

2. Alcohol Curve

In the event that the FC10 detects alcohol, the corresponding level is graphed displayed prior to the result.

3. Test Results

Following display of the alcohol graph, the test result is displayed. The result remains visible on the screen until either the Function or Power button is pressed.
